It is essentially natural defense, ie. for every so many CON points, you will gain an extra DEF point

How many points? Is it a static or variable amount of CON points for 1 DEF point? Does it have any effect other than eventually increasing DEF?
On the topic of stats, what does Luck do? I assume it affects drops and crit rate, but does it do anything else? Also, if my assumption is fully correct, how much does it affect the crit rate? What is the base crit rate, anyway?

It's based off a formula so its variable.
Luck affects crits and drop rates, correct. I can't remember off the top of my head what the base rate is, will take a look.
